Huh. For some reason i had it in my head that AS strings were immutable.

then ...

set fullPath to "G5:bigfolder:2 color:myfolder:image1.eps"
set AppleScript's text item delimiters to {":"}
set text item 3 of fullPath to "CMYK images"

If you need to search for and test the text item, you might want to ...

set AppleScript's text item delimiters to {":"}
then get a list of path items;
determine which item to change;
change it;
convert list of path items back to a full path.
