Supposedly there is a new command line utility that replaces the NetInfo
manager that will allow you to do this.
"dscl" is the utility you want to use.
To learn more type "man dscl". I don't have a Leopard machine in front of
me to give you the exact commands.
Using a combination of dscl and removing the .AppleSetupDone file should
result in the desired removal of the 501 user account, and forcing the Apple
Setup Assistant to run at next startup.
The root account is enabled in the new "Directory Services" application.
