It does, but each line of the rule is probably being executed by a different
shell so your changes are being thrown away. Try:

manual.pdf:      manual.tex
                  export PATH="$(PATH):/sw/bin" \
                  echo $(PATH) \
                  latexmk -pdf manual

Or even, of course:

manual.pdf:      manual.tex
                  PATH="$(PATH):/sw/bin" latexmk -pdf manual
