Will I be able to simply set an automount folder on the new volume and tell Workgroup manager that that is where the user folders will reside or do the user folders need to be located on the same volume as the operating system?

Generally I avoid putting any user data on the OS volume. You should be able to define a new automount on the new drive and then define your new users to live there. Moving the home folders can be problematic as there are some applications that keep absolute paths in their prefs. You can do it, but some stuff might be a bit funky.
