Set up your default user then run this command:

sudo ditto -rsrcFork /Users/defaultuser/ /System/Library/User\ Template/English.proj/

this will copy the default users set up to the user template that will then be used whenever a new user is created.

What settings are you trying to change? A lot can be done with Workgroup Manager, Apple Remote Desktop and custom install packages...
