chflags uchg <name of file>

Will lock the file so that only root can unlock it.

chflags schg <name of file>

Will lock the file so that it can only be unlocked IN SINGLE USER MODE - be careful with this one!

Use "ls -lo" to view the flags on a file, nouchg or noschg to unlock it.
