If the backups were made while the servers were in target disk mode,
there should be no issues with locked/unreadable files.
I think the core concern is that something is missing from one of the
backups. A great way to test this is to write the image back to a
spare machine. Does it boot ? Is there anything obvious missing ?
I'd also do some basic enumerations of the filesystems and compare. On
the server and the corresponding image:
#How many files and folders do we have
find / | wc -l
#What is the sum size of those files and folders
-du -sxk /

As far as I know Mac OS X writes files to disk constantly AND Disk
Utility
doesn backup locked files. But I'm not the expert so I could be wrong.
Others with more experience should say something about this.
