I checked our PPD and found the following:
*OpenUI *Duplex: PickOne
*OrderDependency: 50 AnySetup *Duplex
*DefaultDuplex: DuplexNoTumble
*Duplex None/Simplex: "<</Duplex false /Tumble false >> setpagedevice"
*Duplex DuplexNoTumble/Duplex long-edge binding: "<</Duplex true /Tumble
false>> setpagedevice"
*Duplex DuplexTumble/Duplex short-edge binding: "<</Duplex true /Tumble
true>> setpagedevice"
*?Duplex: "
currentpagedevice dup /Duplex get {
/Tumble get {(DuplexTumble)} {(DuplexNoTumble)} ifelse
}{/Tumble get {(SimplexTumble)} {(None)} ifelse

As you can see the DefaultDuplex is DuplexNoTumble, but this is not
taken over in the standard mac printer driver Layout pane.
I tried to look up what PPD-keyword match with the options in the layout
pane but I did not find anything appropriate on that.
Do you have a suggestion?

BTW it would be marvellous if we could arrange setting the default
duplex value in this way!!!

If it's a CUPS driver, can you just make those defaults in your PPD?
