Right, I just tried it using your path

/System/Library/CoreServices/RemoteManagement/AppleVNCServer.bundle/ Contents/Support/LockScreen.app/Contents/Resources/

and it works.

Earlier with ARD 2 when i tried to replace files inside bundles or packages instead of putting it inside the bundle it created a folder besides the bundle with the bundle's name and placed the file into that folder. So I replaced the entire bundle already containing the new file, but had lots of user rights voodoo later.
