It's Java, but don't be put off by that - the OSX version runs very well.
This is a version of the old NIH Image, which ran on Classic Mac. I don't
know if it will do what you want automatically, but there are a bunch of
plugins, and it does quantitative analysis of images in various ways: counting
particles, for example.
