As was pointed out, this is not a USB question. Other than the Darwin lists, you might also find the carbon-development list helpful:

You didn't say what error code was returned, but usually a volume can't be unmounted because there are files open on the volume. On Mac OS X, the lsof command will list open files.

Also, on Mac OS X, if you have an application with its current working directory on that volume, it won't be ejectable. For example, if you have a Terminal session where you've issued a cd command to some directory on that volume, the volume won't be unmountable.
