Corrupt Keychain on the local user will cause this issue.
Delete local keychain and restart seems to have fixed it.

I've had issues with Configurator too! Prior to the update on January 13th, everything was working fine. Since then, I cannot even get Configurator to OPEN. I'm hesitant to start over, since I'd have to recreate ALL of my settings, profiles, etc. Just me?

I haven't used Configurator for 3 weeks until today. Now, many of the iPads, when hooked up, will not sync. On my laptop, a window appears saying "Unable to attach device to Apple Configurator". Why is it doing this?
Also, on every iPad, it asks me whether I should trust the computer or not, despite the fact that this is the laptop I have always used to add apps to the iPads.

I had the same problem. You should trust the computer and that should fix the configurator issue. I do not know why that happens, but that worked for me.
