I've just updated to Snow Leopard (only OS and no XCode) and my command line make command is no longer available.
It can be found in /Developer/usr/bin but does no longer seem to be in the search path.
Does this indicate that I must also install the new XCode?

Have you disabled the checkbox in the Xcode installer that said something like "install support for UNIX development"?
