It depends.

In reverse order:

> have it respond with a comment?

 say "a comment"

> allow me to say a command to the TTS engine

Save it as a script application in ~/Library/Speech/Speakable\ Items with a
file name based on what you want to say in order to invoke the script. If
you name it "a command" then it will run the script any time the speech
recognition engine hears that phrase.
