Prior to Leopard, you could do that via an applescript, but scripting signatures broke in Leopard, and has never been fixed. I have only 150 in my sig file; the initial 100 or so were inserted with that script, and the rest were hand entered over a period of several years.
The best you can do now with a script is append the "signature" to the body of the message.
