At the risk of sounding unfashionable, I reckon that's a waste of your time. Do it when you get a problem, by all means, but if you don't have a problem, it's pointless.

I found, accidentally, that it is worth trying for problems where you might never consider a safe boot. My 10 month old iMac became unable to read CDs. My superdrive had already been replaced under warranty once for a similar problem. Of course! It had to be the drive. The day before my AppleCare appt. I created a script applet that quit all apps and ordered a shutdown. That was dumb as my Mac would startup and immediately shut down after running the applet due to the resume “feature.”

I did a safe boot to remove the applet and restarted. Before my Genius bar appt. I checked to make sure things were really doing what I was going to describe to the genius, and I no longer had a superdrive problem. The safe boot somehow fixed the superdrive problem.
