You mean content indexing as in the file contents itself? Thats done at save time via spotlight plugins, or a forced re-index (refresh) of the spotlight database, again using spotlight plugins.
If you mean initiating a content index actively (not passively letting spotlight do it when it wants to) then you can probably touch the file to cause an automatic re-scan by spotlight or utilize mdimport (http://developer.apple.com/documentation/Darwin/Reference/ Manpages/man1/mdimport.1.html) I believe.
