URL Connection block?

I would like to make an application that blocks specified URL connection
system-wide.
It may work somewhat similar to Little Snitch.
For example, if Safari tries to connect to a specified URL, for example,
http://www.example.com
then it will block the connection and inform a user about it through pop up
window or something.
However, I don't know how to block specific connection system-wide.
Should I do research on kernel for this?
