When dealing with applications (application bundles) by name or path,

Front Row

This seems to be pretty important, and I'm curious when and to what
degree I should be cautious about the naming used in code.

What are you really trying to do? If you're trying to access the package as a directory, then of course you need to use the real name. If you're trying to do something else with it...well it's entirely possible you shouldn't care about "the naming used in code" at all. It would, for example, not be a best practice to hard-code the string "TextEdit.app" into your program.
