Are there profiling improvements - upgrading from PrintOpen 3.0.1 to the
current version of 4?

a. Same separation parameters now apply to perceptual and relative colorimetric.

b. Higher GCR for high-speed printing.

c. Support for 16 bit profiles which reduce the impact of CMM white point oddities. Use the 'Large' setting for this. Proofer profiles don't travel as attachments and don't get embedded on an object-specific basis in PDF the way press profiles do. So why save on space for a single, stationary profile.

d. White point editing in all three channels in addition to the existing feature that only modifies the top of the L channel in the proof profile. Use white point modification if you get a press / proof combination where the paper white needs a massage. You can have an OK CMM and two OK profiles and still come away with a paper white mismatch, either because the proofer doesn't have a stable first printing point or because the given profile pair doesn't lead to a good interpolation result. In this case change the a and/or b channels of the proofer profile, but leave the press profile alone for practical reasons.

Some UCR enhancements under the hood improve control of dark colors, again for high-speed printing.

There are some bells and whistles in the PC UI, but a copy of ColorThink should be a cure for that.
