Andy,
The simple answer is that a colorimeter uses colored filters to simulate
the response of the Standard Observer, whereas a spectrophotometer
measures spectral power (typically in 10, 5 or 1 nM increments in
wavelength) and from there on either it or you calculate tristimulus
values mathematically.
