profiling help

We have created profiles of a printers output device using Monaco
Profiler. Do we now use that profile to create our separation for photos
that will be printed on their output device? If so, do I soft proof it
in Photoshop using that profile? Is that profile also used in page
layout programs like QuarkXPress? Does the profile have to be sent along
when the job gets output?
Also, on our Iris proofer I have the ability to set the Iris profile
for the Iris and set a target profile to make the printer mimic another
press. So if I sent down a file that has been tagged in Photoshop for
one press and set the destination for a different press, will the
printout show me what the file should look like on the destination
press, or would it just give me unreliable colour that resembles
nothing?
