Basically yes. High end (or low) PMT and filters will 'see' each film
differently and would require different profiles, but you can get away with
1 profile if you don't mind making a samll tweak to the scan in Photoshop.
Or tweak the existing profile for the other material.

Most profiling software requires a minimum size file, roughly the size of a
4x5 at 100% so a 35mm would have to be scanned at at about 400%. If you
have the hardware to do it, you can measure the 4x5 target to get a slightly
more accurate profile.

AFAIK, relcol is used mostly when converting a profiled scan to a
work/storage space.
