You can profile on top of ACR as you're simply fingerprinting the capture
mechanism just like you're doing with any other type of profile process.

Actually, that's a very effective way of profiling an Epson in situations
where the No Color Adjustment behavior is so out of wack (non linear) that
you're willing to sacrifice color gamut of resulting profile for better ink
delivery.
You can profile anything pretty much as long as the behavior of what you are
profiling remains consistent which is the case with ACR or the Epson
example. This isn't to say I recommend profiling on top of ACR but it can be
done.
