The reason for measuring the IT8 chart on a lightbox is to create the
reference file for making the scanner profile. This reference file is
input to the profile creation software, along with the image from the
scanner, then it is crunched to make the profile.

The monitor measurement fixture helps block ambient light from
affecting the measurement. If you are measuring with the room lights
on then it would be a good idea to leave the fixture attached for
your
transparency measurements. The alternative is to turn off the room
lights and make sure there is no window or skylight illumination.
The difficulty keeping the monitor fixture attached for the
transparency measurements is that it makes positioning the
spectrometer on the transparency more difficult. Measuring large area
items such as filters would present no problem, but measuring an IT8
transparency is much more difficult with the monitor fixture
attached.

The item being measured here is a lightbox, not a monitor, so turning
off the room lights is appropriate if the monitor adaptor is not
being
used.
On your point of measuring monitors with or without the room lights
on, I have always measured monitors (CRT and LCD) with the room
lights
off and never had any cast issues.
One important point, if there are any other light sources on in the
room that cannot be turned off (such as equipment power indicators or
other monitors, etc.), I cover the item being measured with a black,
light blocking cloth.
