Channel Maps and Stuff

This is a little outside the scope of this list, but I don't know where else to go and I'm sure some people here will have simple answers... I'm trying to support a broad range of audio input hardware, like the MOTU 828 or the M-Audio Delta66. I'm a freeware developer, and have no such devices of my own to test, and I have no idea how to properly read from these devices.

The app is a relatively simple tuner, so I only need a single mono channel. Are there specific channels on these devices that will provide such data? Should I leave it up to the user to select the channel to be read? Are all 8 channels on these devices usable for instrumental input?

I've read the earlier posts regarding channel maps and I believe I have it figured out, so only short answers to these questions, or perhaps any general advice, is necessary/desired.
