Right, so I'm trying multiple instances of AUSampler, connected to a
MultiChannelMixer, connected to output unit, but I don't get any sound,
even with only a single sampler.

I don't get any errors, but no sound. Do I need to enable the inputs on
the mixer some how, or set gain/levels?
It works fine if I connect the sampler directly to the output unit,
bypassing the mixer.
