I'll let others chime in here, but want to throw in my $.02. First, if at all possible, hold this off until school is out for the year. That will allow you to do the testing and migration at your pace without a weekend under the gun. Secondly, I would say that there's a 50/50 chance that this will "just work" when doing an upgrade from 10.5 to 10.6 server. The mappings are different, the supported authentication algorithms are slightly different, and it all depends on what you have enabled/disabled on your AD server. When doing any Mac OS X Server upgrade, I always do a clean install to keep the process fresh in my brain. If you've got a full clone or image of the server before the upgrade, as well as an OD archive, you can march boldly into this process once the kiddos leave for the summer. Anybody want to share there war stories here?
