if you updated today, it may have something to do with the Ajax framework update Mike did yesterday. The framework now supports load- on-demand for Javascript dependencies.
Maybe there are some side effects with the in place editor?
Doubtful ... It didn't go into Wonder until the 4am build this morning, but regardless, I just setup this same scenario in the AjaxExample app and it works fine. I have an AjaxUpdateContainer surrounding 3 AjaxInPlaceEditors and an AjaxUpdateLink. Everything works as expected -- editing before the update, editing after the update -- in Safari 3, FireFox 2, and IE6. So there must be something else going on ... Load your example app into FireFox and run it with FireBug and see if there are any errors coming out.
