No. MacPorts installs its own version of libraries rather than using system libraries. This is true for pretty much all libs, not just X11.

Well if you notice, the maintainer of X11 for MacPorts is the same person as the maintainer of the XQuartz project (me), so you're fairly safe using both at the same time... I do.
