In my experience, at least, if you have a single localization you should use only the short form: en, pt, fr, de, etc. Otherwise, if you have (say) only pt_BR, users from Portugal that have set their systems to "generic" Portuguese will fall back to English.

In other words, use pt_BR if you _also_ have a separate pt_PT localization; if you have only a single localization that might be acceptable to all Portuguese-speakers, use pt.
