I don't think Xcode 4.3 runs on Mountain Lion, so the answer is that you can't really do this. If you hunt the developer forums you'll find someone who posted the steps required to rip the iOS4.3 simulator out of an old Xcode and run it on ML but I have no idea if it really works or not.
In general having more than one Xcode seems to be ok for me, I often have a few versions lying around. Since they are now all packed in a single bundle in Applications they just seem to 'work'. There is probably some issue I'm going to run into with command line tools or similar which is out of the package, so I always keep those up to date with the latest released version, has worked for me thus far.
