It does take a while for Spotlight to index a drive. Traditionally, the spotlight icon in the top right will have a "dot" in the middle of the magnifying glass.

I've been experimenting this morning. I created a 4.7 GB sparsebundle and filled it with more than 830 MB of email files. I tried to do a Spotlight search on this disk, but, as expected, found nothing because the disk is not indexed.
In terminal, the command
mdutil -i volumepath
enables indexing and I can now search and find within the disk.
I was able to search immediately after enabling indexing. There was no wait while indexing was completed. I am suspicious. Comments about Spotlight I have read in the past indicate that indexing may take some time.
Perhaps a Spotlight expert can explain more about what is happening here.
