I have a similar script that changes my signature to the currently playing track in iTunes. I made it a stay-running script that wakes up every couple of minutes, checks to see if what's playing in iTunes has changed and if so, sets the signature in Mail and then sleeps. I figure that I can't write emails that fast anyway, so a couple of minutes delay for a new signature is a reasonable delay…
Of course, I leave typically leave Mail running all the time, but the script does make sure that both Mail and iTunes are running before it does anything.
I was poking about the prefs in Mail (in Lion) just now and found, that like iOS, you can have automatically CC yourself on outgoing email. You could set this option and then create a rule that triggers your script when you receive a message from yourself and then deletes the message.
