Once-upon-a-time, I, too, looked for ways to write Spotlight metadata ... and found nothing.
Today, I tried again by Googling "metadata write mdls". I didn't find what I was looking for.
But, I did find these, which look merely interesting ...

Spotlight metadata contains Finder metadata such as Comment, Creation Date, Modification Date, and others, so you can write Spotlight metadata via the Finder. But, that isn't what you want.
