Trash iTunes completely and download a whole new copy from Apple here:
http://www.apple.com/itunes/download/
If that still happens then repair permissions... the only difference I
have noticed with 4.2 is that it is much speedier and that you can open
the Music Store in a new window by double clicking the Music Store
button.
