There is no great mystery about this. Measure the samples in a
way that best represents what your intended users will see.
If in general they will be looking at at one page in a stack,
measure a stack. If they will fasten a sheet to a black
surface, measure on a black surface. If they will fasten it
to a grey surface, measure on a grey surface. etc. etc.
There may be no one answer, but decide what you're assumptions
are, and measure based on that assumption.
