They're completely different colorspaces and different gamut shapes, but Proof Setup is showing you a preview of the conversion to CMYK, not the differences between the color spaces. The fact that you see no difference between the RGB and the CMYK preview shows that 1) the colors in the image are more or less already in within the CMYK gamut, and 2)your profile is fairly accurate. If you get some AdobeRGB images with a little more saturation, you very well may see some shifting in the CMYK preview (add a Hue/Saturation layer and crank the saturation slider and you see a difference). I usually see shifting in the black  point, blues and yellows fist, but of course this all depends on the image.
