You can't actually calibrate to D65 (any more than you can 6500K). But when
a software product offers BOTH options (and many do), I advise users always
pick the D illuminant setting over a CCT setting. We have no idea under the
hood what is actually happening here. Since we are dealing with CCT, which
is a range of color versus asking for a specific color (an illuminant), it's
safer to select that when given the choice.
Few software products allow specification over xy values. If they do, then
use that of course. If not, use a D Illuminant. It's just less ambiguous.
