Armand,

Anything that affects appearance upon output needs to be included in the
measurements from which one builds an output profile.

The gloss optimizer does affect the appearance of the printed image --
however slightly. Also, any particular combination of ink and paper on a
specific inkjet printer will produce a different output as well (if that is
what you mean by "each setting").

Therefore, all of these factors will be part of what is being measured via
the spectrophotometer, because they all affect the final appearance of the
printed image.
