The item being measured here is a lightbox, not a monitor, so turning
off the room lights is appropriate if the monitor adaptor is not being
used.
On your point of measuring monitors with or without the room lights
on, I have always measured monitors (CRT and LCD) with the room lights
off and never had any cast issues.
One important point, if there are any other light sources on in the
room that cannot be turned off (such as equipment power indicators or
other monitors, etc.), I cover the item being measured with a black,
light blocking cloth.

Are you sure?
I have seen others, and my own, profiled monitors with lights off and
can see a hard cyan cast.
