You can use XRite's MeasureTool to do exactly what you describe.
First, you must download the ProfileMaker installer from this URL:

Since you seem not to have a license, some of the ProfileMaker products will not work for you. But you will still be able to use MeasureTool to do what you are describing.
Once you have installed ProfileMaker, launch MeasureTool. Unpon launching, it will prompt you with a message that reads: "This application will run in a limited demonstration mode. A hardware dongle is required to activate all functions." That comes up because you don't have a ProfileMaker dongle. Click OK.
Go to Tools > Instrument Configuration. Make sure your EyeOne Pro is plugged in to your computer. Choose "EyeOne Pro" from the "Instrument" pulldown menu. If MeasureTool is able to "see" the plugged-in EyeOne Pro, it will show an "OK" in its lower left-hand corner.
Close the "Instrument Configuration" dialog box.
Go to Tools > Spot Measurement. You will be asked to place the EyeOne Pro in its cradle (the one with the small ceramic tile in it: make sure the ceramic tile is clean, but don't touch it! Just carefully blow at it with an air can). Click OK.
In the "Spot Measurement" dialog box, make sure that the "Color Space" is Lab, the "Observer angle" is 2 degrees, and the "Illumination" is D50. Then place the EyeOne Pro on top of your specimen, then take your reading (place a few white sheets of paper underneath the specimen if it's a thin piece of paper).
The Lab values will appear first next to "Reference", then next to "Sample" upon subsequent readings.
