Unfortunately, no and no.

MIDIServer doesn't do anything in response to a SIGHUP nor is there a nice way to get it to re-evaluate the driver plugins.

For now you should probably instruct the user to quit all applications which might be using MIDIServer.

You could file an enhancement request for a way to tell MIDIServer to look for new or changed drivers and reload them.
