This is of course what tools such as Csound do falling off a log. The
"CSD" file format (plain text) contains a score defining instruments
(which may be as simple as a sample player opcode), and the score part
defines the note list. You can define samples to play in either. It
supports both batch performance and real-time/interactive. LGPL though,
which may not suit. Or there is STK (simple C++), which also supports a
scoring system and has no licence restriction. But it does all involve a
bit of programming/scripting, one way or another.
