So it definitely seems that I can only reproduce this issue when I load my caf files directly rather than from an aupreset - however if I save out an aupreset file in both scenarios the preset file has the voice count set to 32, and stranger still if I load
the aupreset file generated after loading the caf files I still hear the problem. At this point I'm about ready to give up and struggle on using AULab...

Interesting – I think I am going to need to dig a bit deeper to figure out what exactly is going on.
This is on iOS – previously I had been using an aupreset and hadn’t noticed any problems with polyphony (since 32 is
probably high most of the time in my app) however last night I noticed voices cutting out and assumed it was related to my switching away from using aupresets – however it is possible that I just never had a case before that revealed the problem.
I will have a bit more of a dig tonight and see if it is specific to not using aupreset files or across the board.

As far as I can see, and searched around, using various parameter/property list polling, and all available documentation, and feedback from Core Audio engineers in Apple's dev forums, there seems to be no programmatic way to do this..
If we are talking about iOS, does the .aupreset method work for you? Because when I tried, I had no luck overriding what seemed to be the default polyphony of 32 (if I remember correctly) by .aupreset. I tried this by changing the polyphony
in AU Lab (indeed the only user-facing way it seems) and exported as an .aupreset, or manually "tweaking" the .aupreset in a text editor. I would then assign the .aupreset to the running AUSampler, but it would not seem to have any effect on polyphony (notes
cut off at the same time), and indeed when I re-exported the .aupreset out of the running AUSampler, it was still at its default polyphony unchanged.
