I've just updated to Snow Leopard (only OS and no XCode) and my command line make command is no longer available.
It can be found in /Developer/usr/bin but does no longer seem to be in the search path.
Does this indicate that I must also install the new XCode?

Yes, installing Snow Leopard over Leopard removes all of Leopard's / usr/bin developer tools, and you need to install them from the Xcode installer disk image on the Snow Leopard DVD.
