it would seem that it's:

com.apple.internetconfig.plist (Version 2.5.3/ic-added/Helper:http and Helper:https)

however the value is binary (perhaps it's a file reference),
and i recall hearing about some interesting difficulties of
people who were trying to work around the behavior of
Explorer constantly regaining default status ... i just
tried switching my pref to OmniWeb and the value in that
plist did not change, so perhaps it is also cached until
logout.

further, when i made that change, the files which control
application binding (LSSchemes, LSClaimedTypes &
LSApplications) had their mod dates bumped. i don't believe
the format of those files is publicly specified ...

also note that if you edit a plist for some aspect of the
system that is normally always running, your changes may be
overridden at logout unless you can figure out a way to
signal the relevant component to reread its prefs ...
