Is it worth or necessary to upgrade from PrintOpen 3.1 to 4.0.
Any major improvements besides technical stuff like USB support?
Same question for ViewOpen ICC 1.0 to the new 4.0 version

PO4 has white point editing (after you calculate the profile), and the same black generation is applied to all LUTs. The downside is that you have to know how to profile a wet ink process now, at least in the Mac version which doesn't have sample profile presets, a feature introduced in PO3.1 PC, and also available in PO4PC. Under the hood there are many improvements in the algorithms. For instance, GCR can now be run up to 90% for newsprint.

VO4 Mac / PC supports the Apple VCGT tag, builds both matrix-based monitor profiles for Pshop 5 and LUT-based for Linocolor / Newcolor and upgraded Adobe tools, and has some other enhancements.
