We had a problem that sounds similar, but you'll have to give me more
information (off-list) about the term "darken".
You may also need to resist temptation to lighten the shadows of 16-bit RGB
tranny scans. They look as if all shadow detail has been lost when you view
them on your monitor - but hitting the "equalize" button will reveal masses
of information.

That's easy.
Open up your freshly scanned target in Photoshop and either:
A. apply a curve to *darken* the image, or
B. adjust the grey slider in levels to do the same
Save your new "darker" target and then create a profile using for favourite
profiling software.
This seems to work OK for images that are destined for CMYK and are using
the same CMYK profiles that we would have used when our scanner operators
were making corrections visually.
It also seems to work better for operators who are used to seeing more open
shadow detail...
