scanner profiling question

How do you profile a scanner like the Nikon 4000 or 8000, which can give 14 bit TIFF files? Do you make a scan of the film target in 14 bits and convert to 8 bits in PS before going into the profile generation software?

I did find the setting that turns off auto-exposure in the Nikon 3.1 software. I assume that if I also turn off "Nikon Color Mngmnt" (whatever that is) then the scanner is locked down and ready to be profiled, right...?

Also, are the analog gain settings on these scanners useful in some situations? If so, when?
